SCR-M015 is a bispecific antibody designed to simultaneously target programmed cell death protein 1 (PD-1) and vascular endothelial growth factor A (VEGF-A). Developed by Kelun-Biotech, the molecule features a 2+2 symmetrical structure created by fusing an anti-PD-1 nanobody to the C-terminus of the heavy chain of an anti-VEGF-A antibody, which incorporates a silent Fc region to minimize effector-mediated toxicity. This dual-action mechanism aims to provide synergistic anti-tumor effects by blocking the PD-1/PD-L1 immune checkpoint to restore T-cell activity while concurrently inhibiting VEGF-mediated angiogenesis and immune suppression within the tumor microenvironment. Preclinical data presented at AACR 2026 demonstrated that SCR-M015 exhibits high binding affinity for both targets and superior anti-tumor efficacy in various xenograft models compared to benchmark monotherapies and combination treatments.
