scrambled-CyAl5 is a near-infrared fluorescence (NIRF) imaging probe used as a negative control in the study of atherosclerotic plaque erosion. It consists of a scrambled peptide sequence conjugated to the CyAl5 fluorophore (excitation/emission 643/661 nm). Unlike its targeted counterpart, HAT-CyAl5, scrambled-CyAl5 lacks specific binding affinity for hyaluronic acid (HA), a key biomarker enriched at the luminal interface of eroded plaques. In experimental models, including electrical injury-based mouse models and human carotid specimens, it is used to establish baseline fluorescence and demonstrate the target-specific uptake of hyaluronan-targeted agents. The probe was developed by researchers at Massachusetts General Hospital and the Masonic Medical Research Institute.
