Scrambled OPN peptide 1 is an experimental control peptide used in preclinical research to evaluate the efficacy of active immunotherapies targeting Osteopontin (OPN). It consists of a randomized (scrambled) amino acid sequence derived from a functional OPN epitope, such as the integrin-binding RGD motif. In studies investigating cardio-metabolic diseases, this peptide serves as a negative control to distinguish between the sequence-specific effects of OPN-targeted vaccines and non-specific immune responses. Research published in 2016 (PMID 27639826) utilized this peptide in Ldlr-/- and wild-type mouse models to demonstrate that while OPN is a key cytokine in obesity-linked inflammation and atherosclerosis, peptide-based vaccination strategies faced significant challenges, including a failure to induce antibodies capable of binding the endogenous functional protein.
