sCSF-1RWT-Fc is a recombinant fusion protein consisting of the extracellular domains (D1 to D5) of the wild-type human colony stimulating factor-1 receptor (CSF-1R) fused to the Fc region of a silenced human IgG1. It functions as a decoy receptor or "trap" that binds and neutralizes the CSF-1R ligands, colony stimulating factor-1 (CSF-1) and interleukin-34 (IL-34). By sequestering these ligands, it inhibits CSF-1R signaling, which is involved in the recruitment and differentiation of immunosuppressive tumor-associated macrophages (TAMs) and the proliferation of certain tumor cells. In preclinical studies, particularly in pleural mesothelioma models, it has been used as a wild-type control to demonstrate the therapeutic potential of blocking the CSF-1/IL-34/CSF-1R axis and to evaluate the enhanced efficacy of mutated variants like sCSF-1RM149K-Fc.
