SCY-247 is a second-generation triterpenoid antifungal compound, belonging to the novel "fungerp" class of structurally distinct glucan synthase inhibitors. It is being developed by Scynexis for the treatment and prevention of invasive and drug-resistant fungal infections, including those caused by Candida auris and other multidrug-resistant fungi. SCY-247 exhibits broad-spectrum fungicidal activity against yeasts, molds, pneumocystis, dimorphic fungi, and strains resistant to azoles and echinocandins. Its mechanism involves inhibition of 1,3-beta-D-glucan synthase—an enzyme essential for fungal cell wall synthesis—resulting in disruption of cell wall integrity and ultimately fungal cell death. The compound demonstrates high tissue penetration and favorable tolerability in preclinical models. As of June 2025, SCY-247 is in Phase 1 clinical trials evaluating safety, tolerability, pharmacokinetics (oral formulation), with both oral and intravenous formulations under development[1][2][4][5][6][10].
