Drug intelligence / Profile preview
SD-133 is a drug-like small molecule inhibitor of cadherin-11 (CDH11) originally discovered and developed by researchers at Georgetown University. It binds specifically to the homodimerization interface (the W-binding pocket) of CDH11, preventing homophilic cell-cell adhesion. SD-133 has been investigated in preclinical models for its therapeutic potential in various cancers, including breast cancer, pancreatic cancer, and adenoid cystic carcinoma, where CDH11 overexpression drives tumor progression, invasion, and metastasis. Additionally, SD-133 has demonstrated nephroprotective properties in preclinical models of chronic kidney disease by reducing renal fibrosis and inhibiting the pro-fibrotic SMAD3 pathway.
