SD-254 is a deuterium-substituted analog of venlafaxine, a serotonin-norepinephrine reuptake inhibitor (SNRI), designed to improve pharmacokinetics by slowing metabolism through the kinetic isotope effect. Developed initially by Auspex Pharmaceuticals (later acquired by Teva), it demonstrated in Phase I trials a metabolism rate half that of venlafaxine, resulting in longer persistence at effective levels in the body and potentially fewer side effects.[1][5][11] Primarily targeted for major depressive disorder, it also showed promise for neuropathic pain, an indication not formally pursued with venlafaxine.[1][11]
