sD-NP-GT8 + IL-12 DNA + Trimer 4571 is an experimental HIV-1 vaccine regimen developed by the National Institute of Allergy and Infectious Diseases (NIAID). The regimen employs a germline-targeting strategy designed to induce broadly neutralizing antibodies (bNAbs) of the VRC01 class. It consists of a priming phase with synthetic DNA (sD) plasmids encoding the NP-GT8 immunogen (a multimeric protein displaying the VRC01-binding site) and Interleukin-12 (IL-12) as a genetic adjuvant. This is followed by a boosting phase with a stabilized recombinant HIV-1 envelope trimer, Trimer 4571, formulated with the TLR7/8 agonist adjuvant 3M-052-AF and alum. The combination is currently being evaluated in Phase 1 clinical trials (e.g., HVTN 305) for the prevention of HIV infection in healthy adults.
