sD-NP-GT8 DNA is a synthetic DNA vaccine candidate designed to encode a germline-targeting HIV immunogen, specifically the GT8 epitope, which is engineered to stimulate broadly neutralizing antibody (bNAb) precursors against HIV. The vaccine utilizes a nanoparticle-based approach, where the GT8 immunogen is scaffolded onto nanoparticles and delivered via plasmid DNA for in vivo expression. This strategy aims to prime B cells capable of maturing into bNAb-producing cells. The vaccine has been evaluated in combination with IL-12 DNA as an adjuvant and may be followed by protein boosts such as trimeric Env proteins (e.g., Trimer 4571). It is being developed primarily for HIV prevention and has been tested in early-phase clinical trials involving healthy adults without HIV[1][2][3][5].
