SD55-TSLC1 is a survivin-regulated oncolytic adenovirus armed with the TSLC1 gene (tumor suppressor in lung cancer-1/cell adhesion molecule 1) developed for cancer therapy. Its mechanism targets cancer cells via two main actions: agonism of CADM1 (cell adhesion molecule 1, also known as TSLC1) and inhibition of survivin (baculoviral IAP repeat-containing protein 5). The selectivity comes from the survivin promoter, which is more active in cancer cells (particularly hepatocellular carcinoma) than in normal cells. Upon infection, SD55-TSLC1 overexpresses TSLC1 in tumor cells, activating caspase-mediated apoptosis pathways, leading to selective tumor cell death while sparing normal tissue. Preclinical studies show SD55-TSLC1 inhibits liver cancer growth both in vitro and in vivo. The drug was developed by Zhejiang Sci-Tech University for neoplasms, mainly hepatocellular carcinoma, and is currently at the preclinical development stage[1][3][5].
