Overview

sdAb10 CAR-T is a preclinical, autologous chimeric antigen receptor (CAR) T-cell therapy designed to target B-cell maturation antigen (BCMA) for the treatment of relapsed or refractory multiple myeloma (R/R MM). Developed by researchers at Cima Universidad de Navarra and Nanogrow Biotech, the therapy utilizes a single-domain antibody (sdAb), or nanobody, as the extracellular antigen-recognition moiety instead of a traditional single-chain variable fragment (scFv). The CAR construct incorporates a 4-1BB costimulatory domain and a CD3-zeta signaling domain. In preclinical xenograft mouse models presented at ASH 2025, sdAb10 CAR-T demonstrated enhanced antitumoral efficacy and improved survival rates compared to the approved CAR-T therapy idecabtagene vicleucel (ide-cel). The use of sdAbs is intended to reduce tonic signaling while maintaining robust activation and improving the memory phenotype of the engineered T cells.
