Drug intelligence / Profile preview
sdBCMA.BBz CAR-T cells are an investigational chimeric antigen receptor (CAR) T-cell therapy being developed for the treatment of multiple myeloma. The therapy utilizes a single-domain antibody (sdAb or nanobody) targeting B-cell maturation antigen (BCMA; TNFRSF17) linked to a 4-1BB (BB) co-stimulatory domain and a CD3zeta (z) signaling domain. Developed through a collaboration involving the National Research Council Canada, BC Cancer Research Institute, the University of Calgary, and the University of California, San Francisco, these CAR-T cells are designed to provide effective anti-tumor activity against BCMA-expressing multiple myeloma cells. To address the risk of cytokine release syndrome (CRS) and inhibit IL-6-driven myeloma growth, researchers have also engineered versions of the sdBCMA.BBz construct that incorporate membrane-bound or secreted single-domain antibodies that neutralize interleukin-6 (IL-6). The therapy is currently in preclinical development.
