SDF-061390E is a novel multivalent, multispecific antibody-drug conjugate (ADC) developed by Synthetic Design Lab using their proprietary Synthbody™ platform. It is designed to target three distinct antigens commonly expressed on human myeloma cells: B-cell maturation antigen (BCMA), G protein-coupled receptor class C group 5 member D (GPRC5D), and CD38. The construct incorporates 6-12 tumor antigen binding domains, enabling combinatorial logic-gated targeting. This architecture is intended to replicate high-expression targeting (similar to HER2) even in tumors with heterogeneous or low individual antigen expression. Preclinical data indicates that SDF-061390E (and its monomethyl auristatin F conjugated form, SDF-061390E-MMAF) demonstrates significantly enhanced internalization and potency compared to mono-specific agents like belantamab mafodotin, while also evading the inhibitory effects of soluble BCMA.
