SDF-061390E + MMAF is a multivalent, multispecific antibody-drug conjugate (ADC) developed by Synthetic Design Lab using their proprietary Synthbody™ platform. It is designed to target three distinct antigens commonly expressed on human multiple myeloma cells: B-cell maturation antigen (BCMA), G protein-coupled receptor class C group 5 member D (GPRC5D), and CD38. The Synthbody™ architecture incorporates 6-12 tumor antigen-binding domains, enabling logic-gated targeting and enhanced internalization. The drug is conjugated to the cytotoxic payload monomethyl auristatin F (MMAF), a tubulin inhibitor. In preclinical studies, SDF-061390E + MMAF demonstrated significantly greater potency and internalization compared to the BCMA-targeting ADC belantamab mafodotin, and it remained effective even in the presence of soluble BCMA or in cells with low target expression.
