An SDF1 neutralizing antibody (also known as an anti-CXCL12 antibody) is a therapeutic modality designed to bind and neutralize stromal cell-derived factor 1 (SDF-1, or CXCL12), a homeostatic chemokine that plays a critical role in cell migration, hematopoiesis, angiogenesis, and tumor microenvironment regulation. By blocking the interaction of CXCL12 with its receptors, CXCR4 and ACKR3 (CXCR7), these antibodies inhibit downstream signaling pathways (such as MAPK/ERK and PI3K/Akt) that promote tumor cell survival, metastasis, and immunosuppression. SDF1 neutralizing antibodies are being investigated in preclinical and clinical settings for various indications, including oncology (to disrupt the tumor-supportive niche and enhance immunotherapy), inflammatory diseases, and pathological hair loss conditions like androgenic alopecia and alopecia areata.
