Overview

SDH1 is an engineered strain of the probiotic yeast *Saccharomyces cerevisiae* var. *boulardii* (Sb) that has been genetically modified to eliminate acetate production. Developed by researchers at KU Leuven and Novelyeast, it serves as a critical negative control in the development of next-generation live biotherapeutic products (LBPs) for inflammatory bowel diseases. By comparing SDH1 to high-acetate producing strains (such as SbP and ENT3), researchers demonstrated that acetate production is a primary driver of Sb's anti-inflammatory and barrier-protective effects in preclinical models of ulcerative colitis. In these models, SDH1 failed to attenuate disease activity, weight loss, and histologic inflammation to the same degree as acetate-producing variants, highlighting the importance of metabolic engineering in optimizing probiotic efficacy.
