Overview

SDI-118 is a small molecule drug that acts as a modulator of synaptic vesicle glycoprotein 2A (SV2A) in the brain. It is being developed for its procognitive effects, with the aim of enhancing synaptic efficiency and improving cognitive function. Preclinical studies have shown cognitive-enhancing effects in animal models of cognitive deficit. In first-in-human clinical trials, single oral doses up to 80 mg were well tolerated in healthy male subjects, with mild adverse events such as dizziness and somnolence increasing at higher doses. The drug demonstrated dose-proportional SV2A occupancy in the brain, supporting further clinical exploration for cognitive disorders and neuropsychiatric or neurodegenerative diseases characterized by cognitive impairment. It was originally developed by Syndesi Therapeutics before being acquired by AbbVie. It has also been referenced as ABBV-552 post-acquisition.
