Overview

SDL-M1 is a liver-targeted nanoparticle-based therapeutic developed by SmartDyeLivery GmbH for the treatment of septic cholestasis, a severe liver dysfunction occurring during sepsis. The drug consists of a phosphoinositide 3-kinase (PI3K) inhibitor encapsulated within a functionalized polymer nanoparticle. The nanoparticle surface is modified with a polymethine dye that specifically targets Organic Anion Transporting Polypeptides (OATPs) on hepatocytes, allowing for site-specific delivery and high local concentrations in the liver. By inhibiting PI3K, SDL-M1 aims to restore the expression and activity of hepatobiliary transporters that are typically suppressed in sepsis, thereby facilitating the excretion of bile acids and bilirubin and preventing liver failure.
