Overview

SDP-013 is a potent, selective, and orally bioavailable small molecule antagonist of the somatostatin receptor 2 (SSTR2). It was developed by Speranza Therapeutics, a subsidiary of Centessa Pharmaceuticals, in collaboration with Sosei Heptares (now Nxera Pharma). The drug was primarily investigated for the treatment of hyperinsulinemic hypoglycemia, including congenital hyperinsulinism (HI) and post-bariatric hypoglycemia (PBH). Its mechanism of action involves blocking SSTR2 on pancreatic alpha cells; since SSTR2 activation normally inhibits glucagon release, antagonism of this receptor promotes the secretion of endogenous glucagon to elevate blood glucose levels. In August 2022, Centessa Pharmaceuticals announced the discontinuation of the SDP-013 program following a strategic review of Phase 1 clinical data and portfolio prioritization.
