SDP03923-000-9106 (also known as SDP-LIV1) is a novel, optimized antibody-drug conjugate (ADC) targeting LIV-1 (SLC39A6), a transmembrane zinc transporter protein highly expressed in various solid tumors, including breast and gastric cancers. Developed by Jiangsu Hengrui Pharmaceuticals and its subsidiary Shanghai Shengdi Pharmaceutical, the ADC consists of an in-house developed humanized anti-LIV-1 monoclonal antibody conjugated to a proprietary topoisomerase I inhibitor payload, SHR9265 (a novel exatecan derivative with enhanced liposolubility and cellular permeability), via a cleavable glycine-glycine-phenylalanine-glycine (GGFG) linker. It features an optimized drug-to-antibody ratio (DAR) of 6. In preclinical studies, SDP03923-000-9106 demonstrated robust anti-tumor activity, favorable internalization, and a promising safety profile, supporting its potential clinical development for solid tumors.
