SDT-011 is a topical small-molecule drug candidate designed to prevent and treat skin toxicities caused by epidermal growth factor receptor inhibitor (EGFRi) monoclonal antibodies such as cetuximab and panitumumab. By binding to epidermal growth factor receptor (EGFR) on keratinocytes, SDT-011 sterically interferes with the binding of therapeutic anti-EGFR antibodies, thereby reducing their local receptor occupancy, reactivating EGFR signaling in skin, and mitigating the papulopustular rashes and related cutaneous adverse events that frequently necessitate oncologic dose reductions.[1][9][12][13] Developed by EMRIS Pharma in collaboration with academic partners at Hadassah Medical Center and the Hebrew University of Jerusalem, SDT-011 is formulated as a slow‑release cream/ointment that penetrates hair follicles and sebaceous glands, aiming to block EGFRi activity only at the site of toxicity without affecting systemic anti-tumor efficacy.[1][3][11][12]
