Drug intelligence / Profile preview
sE-cad neutralizing antibody is a preclinical monoclonal antibody designed to target and neutralize soluble E-cadherin (sE-cad) fragments. In colorectal cancer (CRC), matrix stiffness promotes the palmitoylation of E-cadherin via the palmitoyltransferase ZDHHC8, leading to its proteolytic cleavage and the release of sE-cad. These soluble fragments act as immunosuppressive factors by binding to the co-inhibitory receptor KLRG1 on cytotoxic T cells, driving T-cell exhaustion. By neutralizing sE-cad, the antibody blocks its interaction with KLRG1, reversing T-cell inhibition and restoring anti-tumor immunity. Preclinical studies have demonstrated that combining sE-cad neutralizing antibody with anti-PD-1 therapy synergistically inhibits tumor growth in colorectal cancer models.
