SE-CAR-T cells (Super-empowered CAR-T cells) are an engineered chimeric antigen receptor (CAR) T-cell therapy designed to treat CEA-positive solid tumors. Developed by Chongqing Precision Biotech, these cells are modified with a domain-swapped chimeric receptor (DSCR) consisting of the TGF-β receptor extracellular domain and the IL-9 receptor gamma (IL-9Rγ) intracellular domain. This architecture converts the immunosuppressive TGF-β signal within the tumor microenvironment into a proliferative and survival signal by activating the STAT5/IL-9 pathway. Additionally, the cells express membrane-bound IL-2 (mbIL-2) to enhance T-cell fitness, expansion, and persistence. Preclinical studies in various xenograft models, including gastric, rectal, lung, and cecal cancers, have demonstrated superior antitumor efficacy and tolerance to the immunosuppressive tumor microenvironment compared to standard CAR-T cells.
