It is designed to activate innate and adaptive immunity through CD40 engagement, enhancing anti-tumor immune responses. - **Pembrolizumab** (Keytruda) is a humanized monoclonal antibody that acts as an immune checkpoint inhibitor by binding to PD-1 on T cells, blocking its interaction with PD-L1 and PD-L2, thereby restoring T cell-mediated anti-tumor activity[1][3][5]. - **Gemcitabine** is a nucleoside analog and antimetabolite that inhibits DNA synthesis, leading to apoptosis of rapidly dividing cancer cells. - **Nab-paclitaxel** (Abraxane) is an albumin-bound formulation of paclitaxel that disrupts microtubule function and promotes apoptosis; the albumin formulation enhances delivery to tumor cells[2]. This combination aims to integrate immune activation (via CD40 and PD-1 pathways) with cytotoxic chemotherapy for a synergistic anti-tumor effect, and is currently in clinical trials for cancers such as pancreatic cancer.
