Overview

SEA-TGT is an investigational, effector-function enhanced human monoclonal antibody (mAb) that targets TIGIT (T-cell immunoreceptor with Ig and ITIM domains), an inhibitory immune checkpoint receptor expressed on subsets of T cells and NK cells. Developed using proprietary Sugar Engineered Antibody (SEA) technology, SEA-TGT is a nonfucosylated human IgG1 antibody designed to enhance binding to activating Fcγ receptors and reduce binding to inhibitory Fcγ receptors. This modification increases the depletion of TIGIT-positive regulatory T cells, amplifies naïve and memory CD8+ T-cell responses, and enhances innate immune cell activation. Preclinical studies have shown that SEA-TGT elicits superior anti-tumor immune responses compared to other TIGIT antibodies without effector-enhanced backbones. It is being evaluated as monotherapy and in combination with anti-PD-1 agents for advanced solid tumors (including non-small cell lung cancer, gastric carcinoma, cutaneous melanoma, head and neck squamous cell carcinoma, bladder cancer, ovarian cancer or triple-negative breast cancer) as well as lymphomas (classical Hodgkin lymphoma, diffuse large B-cell lymphoma or peripheral T-cell lymphoma)[2][3][4][5][6][7].
