secreted single chain IL-12p70 mRNA is an mRNA-based immunotherapy developed by Geneleap (a subsidiary of Luye Life Sciences) for the treatment of solid tumors, including triple-negative breast cancer (TNBC). The drug consists of a synthetic mRNA transcript that encodes a single-chain version of the IL-12p70 cytokine, which is the bioactive heterodimer composed of p35 and p40 subunits. By using a single-chain design, the mRNA ensures the production of the functional cytokine from a single open reading frame, improving protein bioavailability within the tumor microenvironment. Administered intratumorally, the mRNA is translated by local cells to secrete IL-12, a potent pro-inflammatory cytokine that promotes a Th1-type immune response, activates natural killer (NK) cells, and stimulates cytotoxic T lymphocytes. This localized delivery approach aims to induce robust anti-tumor immunity while minimizing the severe systemic toxicities historically associated with recombinant IL-12 protein administration. Preclinical studies have demonstrated that intratumoral dosing of this mRNA induces significant tumor regression in syngeneic and orthotopic mouse models of TNBC.
