SEL-403 is an investigational combination drug consisting of SVP-Rapamycin (an immune tolerance platform) and LMB-100 (a recombinant fusion immunotoxin targeting mesothelin). The primary mechanism is to reduce the immune response against protein therapeutics, enabling repeated dosing with the immunotoxin for enhanced anti-tumor efficacy. SEL-403 combines SVP-Rapamycin to prevent immunogenicity and LMB-100, which inhibits protein synthesis by targeting mesothelin, a cell surface protein highly expressed in mesothelioma and other cancers. The drug is being developed primarily for malignant mesothelioma that has failed prior chemotherapy[1][10][13]. LMB-100 is derived from a bacterial toxin (Pseudomonas exotoxin A) and directed to cancer cells via an antibody fragment specific for mesothelin, leading to selective cytotoxicity. SVP-Rapamycin allows immune tolerance to LMB-100, permitting multiple treatment cycles and improved safety compared to immunotoxin monotherapy[7][13].
