Drug intelligence / Profile preview
Selcodebart (DISC-0974) is an investigational humanized monoclonal antibody (IgG1 kappa) developed by Disc Medicine, Inc. (originally in-licensed from AbbVie) that targets hemojuvelin (HJV/HFE2/RGMC), a co-receptor in the bone morphogenetic protein (BMP)/SMAD signaling pathway. By blocking the interaction of HJV with BMP ligands, selcodebart suppresses the expression of hepcidin, a key hormone regulating iron homeostasis. This reduction in hepcidin levels mobilizes stored iron and increases iron absorption, thereby enhancing iron availability for erythropoiesis. Selcodebart is primarily being evaluated for the treatment of anemia associated with myelofibrosis (MF), where it has demonstrated significant hematologic responses in the Phase 2 RALLY-MF trial, and is also in Phase 2 development for anemia associated with inflammatory bowel disease (IBD) and Phase 1b development for anemia of chronic kidney disease (CKD).
