**Selcopintide** is a synthetic 10-amino acid peptide (sequence: K.Y.K.Q.K.R.R.S.Y.K) derived from the 344-353 fragment of human Copine7 (CPNE7) protein, developed for dentin hypersensitivity and periodontal regeneration. It mimics CPNE7's role in odontoblast differentiation by binding to nucleolin on pre-odontoblast cells, facilitating internalization, nuclear translocation, and upregulation of odontoblast marker genes such as dentin sialophosphoprotein (DSPP) and Nestin, leading to dentin formation and mineralization. Preclinical studies in human dental pulp cells, mouse pre-odontoblasts, and beagle dog models demonstrated dose-dependent gene upregulation, mineralization, and regeneration of dentin, periodontal ligament, cementum, and bone; a phase 1/2a first-in-human trial showed significant reduction in dentin hypersensitivity (VAS decrease of -23.2 mm at 10 µg/tooth, p<0.05) with excellent safety (no serious AEs, no systemic absorption).[1][2][9][10]
