selgantolimod

Selgantolimod (GS-9688) is an orally bioavailable, selective agonist of Toll-like receptor 8 (TLR8) being developed by Gilead Sciences. It is primarily investigated for the treatment of chronic hepatitis B virus (HBV) infection. By activating TLR8, which is expressed in endosomes of myeloid cells such as monocytes and myeloid dendritic cells, selgantolimod induces the production of pro-inflammatory cytokines (e.g., IL-12, TNF-α) and activates effector cells like natural killer (NK) cells and mucosal-associated invariant T (MAIT) cells. This immune activation aims to restore the exhausted HBV-specific T-cell response and promote the clearance of HBV-infected hepatocytes, potentially leading to a functional cure. It has also been explored in early-phase trials for HIV-1 infection.
