This is an investigational combination therapy consisting of three agents: - **Selinexor** is a first-in-class, orally available selective inhibitor of nuclear export (SINE) that targets exportin 1 (XPO1), leading to the accumulation of tumor suppressor proteins in the nucleus and promoting apoptosis in cancer cells. - **Anti-PD-1 antibody** refers to monoclonal antibodies targeting programmed cell death protein 1 (PD-1), a checkpoint receptor on T cells. By blocking PD-1, these antibodies enhance immune-mediated tumor cell killing. Examples include pembrolizumab, nivolumab, and tislelizumab. - **Golidocitinib** is a highly selective Janus kinase 1 (JAK1) inhibitor that blocks JAK/STAT signaling pathways involved in cytokine signaling and immune regulation. The combination aims to leverage complementary mechanisms—direct cytotoxicity from selinexor, immune activation via PD-1 blockade, and modulation of inflammatory/immune pathways through JAK inhibition—to treat relapsed or refractory cancers such as lymphomas. Clinical studies have evaluated similar combinations for relapsed/refractory primary central nervous system lymphoma and natural killer/T-cell lymphoma[6][2].
