Drug intelligence / Profile preview
This is a combination therapy consisting of four active pharmaceutical ingredients: **selinexor**, **rituximab**, **gemcitabine**, and **oxaliplatin**. Selinexor is an oral selective inhibitor of exportin-1 (XPO1), which leads to nuclear retention of tumor suppressor proteins and is antineoplastic; rituximab is a chimeric monoclonal antibody targeting CD20 on B-cells, mediating cell lysis by several mechanisms including antibody-dependent cell-mediated cytotoxicity; gemcitabine is a nucleoside analog antimetabolite interfering with DNA synthesis; oxaliplatin is a platinum-based chemotherapeutic that forms DNA crosslinks and stalls replication. These agents are combined for the treatment of relapsed or refractory diffuse large B-cell lymphoma (DLBCL) and other B-cell lymphomas in patients who are not candidates for high-dose consolidative therapy or autologous stem cell transplantation. The regimen is under active clinical investigation as a second-line or later option, with early phase clinical data demonstrating tolerable safety and encouraging efficacy profiles[1][3][5][6].
