A combination therapy of selinexor (an XPO1 inhibitor) and venetoclax (a BCL2 inhibitor). Selinexor prevents the nuclear export of tumor suppressor proteins by inhibiting exportin-1 (XPO1), causing their accumulation in the nucleus and promoting cancer cell death. Venetoclax selectively binds to BCL2, blocking its antiapoptotic function and promoting apoptosis. The combination has shown synergistic effects in various hematological malignancies, particularly in multiple myeloma with t(11;14) translocation, acute myeloid leukemia, and mantle cell lymphoma.
