Overview

Selumetinib + dacarbazine is an investigational **combination therapy** for cancer, studied primarily in **metastatic uveal melanoma**. Selumetinib is a potent, selective small-molecule inhibitor of MEK1/2 kinases (mitogen-activated protein kinase kinase 1/2), thus blocking the MAPK/ERK pathway, which is often activated in certain tumor types. Dacarbazine is an alkylating agent and antineoplastic that induces cell death through DNA methylation. This combination was evaluated in the phase III SUMIT trial for patients with metastatic uveal melanoma. The regimen included selumetinib administered orally and dacarbazine intravenously. The combination did not significantly improve progression-free survival compared to placebo plus dacarbazine, although it demonstrated a tolerable safety profile[1][2][3].
