selumetinib + pembrolizumab

Selumetinib + pembrolizumab is an experimental combination regimen consisting of selumetinib, a small molecule MEK1/2 inhibitor, and pembrolizumab, a monoclonal antibody targeting the programmed cell death protein 1 (PD-1). Selumetinib exerts antitumor effects by inhibiting MEK1/2, key kinases in the MAPK/ERK signaling pathway, thereby reducing cell proliferation and survival. Pembrolizumab is an immune checkpoint inhibitor that enhances anti-tumor immunity by blocking the PD-1 receptor, thus preventing its interaction with PD-L1/PD-L2 and reactivating T lymphocyte–mediated tumor destruction. This combination has been investigated for possible synergistic immunomodulatory and anti-tumor activity in patients with advanced or metastatic solid tumors[1][2][3]. The combination is not approved for any indication and remains investigational.
