Drug intelligence / Profile preview
SEMA4A inhibitors are a class of therapeutic agents designed to target and block the activity of Semaphorin 4A (SEMA4A), a transmembrane and secreted protein belonging to the class IV semaphorin family. SEMA4A plays a critical role in immune regulation, angiogenesis, and axonal guidance, and has been implicated in various pathological conditions, including community-acquired pneumonia (CAP), multiple myeloma, and autoimmune diseases. In the context of pneumonia, SEMA4A has been shown to inhibit the transdifferentiation of alveolar type II (ATII) epithelial cells into alveolar type I (ATI) epithelial cells via the AMPK pathway, thereby disrupting alveolar homeostasis and exacerbating lung injury. SEMA4A inhibitors, which may include monoclonal antibodies, small molecules, or other modalities, aim to restore epithelial repair, preserve alveolar homeostasis, and mitigate inflammation, offering a potential therapeutic strategy for lung injury and other SEMA4A-mediated diseases.
