Semaglutide is a glucagon-like peptide 1 receptor agonist (GLP-1 RA) that improves glycemic control and induces weight loss. NNC0385-0434 is a novel oral peptide and selective inhibitor of proprotein convertase subtilisin/kexin type 9 (PCSK9), designed to lower low-density lipoprotein cholesterol (LDL-C). The two drugs are co-formulated with the absorption enhancer SNAC to enable oral bioavailability. This combination targets both glycemic control and cardiovascular risk factors such as hypercholesterolemia. The combination was investigated in a phase 1 clinical study evaluating pharmacokinetics and tolerability in healthy volunteers and is not approved for use.
