Semaxanib + interferon alfa-2b is a combination therapy investigated for the treatment of advanced kidney cancer. Semaxanib (SU5416) is a small molecule receptor tyrosine kinase inhibitor that primarily targets the vascular endothelial growth factor receptors (VEGFR-1, VEGFR-2, and VEGFR-3), thereby inhibiting tumor angiogenesis. Interferon alfa-2b is a recombinant cytokine that exerts direct antiproliferative effects on tumor cells and modulates the host immune response. The combination was evaluated in a Phase II clinical trial sponsored by the City of Hope Medical Center and the California Cancer Consortium to determine if dual inhibition of angiogenesis and immune modulation could improve outcomes in patients with unresectable or metastatic renal cell carcinoma. Development of the semaxanib component was ultimately terminated following disappointing results in other Phase III trials.
