Overview

Semorinemab is a humanized anti-tau IgG4 monoclonal antibody developed as an immunotherapy for Alzheimer's disease. It targets extracellular tau protein, aiming to prevent the proliferation and accumulation of pathological tau aggregates in the brain. The drug was evaluated in multiple Phase II clinical trials (Tauriel and Lauriet) for its potential as a disease-modifying treatment in prodromal-to-mild and mild-to-moderate Alzheimer's disease. While semorinemab demonstrated target engagement—evidenced by increased plasma phosphorylated Tau 181 (pTau181) and cerebrospinal fluid chitinase-3-like protein 1 (YKL-40), suggesting microglial activation—it did not consistently slow clinical decline or cerebral tau accumulation compared to placebo[1][6][7][8]. Semorinemab was developed through a collaboration between AC Immune and Genentech, part of Roche Group[1].
