SEN36 + ricin A chain is an immunotoxin consisting of the monoclonal antibody SEN36 chemically conjugated to the ricin A chain protein. This immunotoxin is designed to selectively target and bind the neural cell adhesion molecule (N-CAM, also known as CD56) on cell surfaces through SEN36, delivering the ricin A chain directly to N-CAM expressing cells. The ricin A chain is a ribosome-inactivating protein (RIP) that enzymatically depurinates a conserved adenosine residue in the 28S rRNA of eukaryotic ribosomes, thereby irreversibly inhibiting protein synthesis and inducing cell death. This construct has been investigated for selective cytotoxicity against N-CAM expressing cancer cells, but evidence indicates that the SEN36-ricin A chain immunotoxin lacked selective cytotoxic activity in tested cell lines[4][1]. Ricin A chain, unlike the complete ricin toxin, lacks the B chain required for broad cell entry, making it dependent on the targeting antibody for cellular delivery and intended specificity[2][1][4]. Primary indications explored for such immunotoxins include N-CAM expressing malignancies.
