Senaparib + temozolomide is an investigational fixed-dose combination of **senaparib** (a novel oral PARP [poly(ADP-ribose) polymerase] inhibitor) and **temozolomide** (an orally available alkylating agent). Senaparib inhibits PARP enzymatic activity and traps PARP at sites of DNA damage, leading to impaired DNA repair in tumor cells. Temozolomide induces DNA damage via methylation. Their combination is designed to promote synthetic lethality: senaparib prevents repair of DNA damage caused by temozolomide, thereby enhancing cytotoxicity. The combination is intended to treat advanced solid tumors and small cell lung cancer (SCLC), with promising results for tolerability and antitumor activity in early clinical trials. The FDA has granted Orphan Drug Designation for this combination in SCLC. IMPACT Therapeutics developed senaparib, and is advancing this combination into global multi-center phase Ib/II trials[1][2][3][5][7].
