Sendai virus vaccine is an experimental live-attenuated, intranasally administered viral vaccine based on the murine parainfluenza virus type 1 (Sendai virus). It is being developed primarily as a Jennerian (xenotropic) live-virus vaccine for the prevention of human parainfluenza virus type 1 (hPIV-1) infection in children. The mechanism of action involves induction of both systemic and mucosal immune responses—specifically increasing serum and nasal IgG/IgA antibodies against hPIV-1—by mimicking natural infection without causing disease in humans. Preclinical studies have shown that it elicits strong protective immunity in animal models without significant adverse effects. Early-phase clinical trials in seropositive children and adults demonstrated that the intranasal administration was well tolerated with only mild transient side effects and no serious adverse events. The elevated antibody levels persisted for at least six months post-vaccination[1][2][6][8]. The Sendai virus platform is also being explored as a vector backbone for vaccines against other respiratory viruses such as RSV and hMPV[2].
