Overview

Seneca Valley Virus (SVV-001) is a naturally occurring, replication-competent, non-enveloped oncolytic RNA virus belonging to the Picornaviridae family. Discovered as a contaminant in laboratory cell cultures, it belongs to the Senecavirus genus. SVV-001 exhibits a highly selective tropism for cancer cells with neuroendocrine features, including small cell lung cancer (SCLC) and various neuroendocrine tumors. Developed by Neotropix, the virus functions as an oncolytic biologic agent that enters cells via the Anthrax toxin receptor 1 (ANTXR1), also known as Tumor Endothelial Marker 8 (TEM8), which is frequently overexpressed in these tumor types. Once inside, the virus replicates selectively within the tumor cells, leading to cell lysis and the release of viral progeny and tumor antigens, potentially stimulating a systemic anti-tumor immune response. Clinical trials have evaluated its safety and efficacy via intravenous administration, demonstrating a favorable safety profile with no dose-limiting toxicities observed in early-phase studies.
