The senescent whole-cell vaccine (SWC vaccine) is an autologous cancer vaccine platform that utilizes conditionally reprogrammed primary tumor cells (CRC method) to enable large-scale in vitro expansion of patient-derived material. These cells are subsequently induced into a state of cellular senescence via gamma-irradiation, creating a highly immunogenic, poly-antigenic substrate. The vaccine is designed to prime systemic T-cell immunity, specifically activating CD4+ and CD8+ effector T cells, and is typically administered alongside a CpG adjuvant. Preclinical studies in murine breast cancer models indicate that the SWC vaccine can break immune tolerance, synergize with immune checkpoint inhibitors (such as anti-PD-L1 and anti-CTLA-4), and establish long-term immune memory to prevent tumor recurrence.
