Overview

Senexin B (BCD-115) is an orally bioavailable small molecule inhibitor of cyclin-dependent kinases 8 and 19 (CDK8/19), which are components of the Mediator complex involved in transcriptional regulation. By inhibiting CDK8/19, Senexin B blocks oncogenic signaling pathways mediated by these kinases, suppresses selective transcription of tumor-promoting genes, and inhibits proliferation in tumor cells that overexpress CDK8/19. It was developed for use in advanced estrogen receptor-positive (ER+) breast cancer, particularly in combination with endocrine therapies such as aromatase inhibitors or fulvestrant. The drug reached phase I clinical trials but development for breast cancer has been discontinued[1][2][3][4][5][8].
