Overview

Senicapoc is a small molecule inhibitor of the Gardos channel (also known as KCa3.1 or KCNN4), a calcium‐activated potassium channel found in red blood cells and other tissues. By blocking this channel, senicapoc prevents the efflux of potassium and water from red blood cells, thereby reducing cell dehydration—a key factor in sickle cell disease pathophysiology. Senicapoc has been investigated for several indications including sickle cell disease, asthma, interstitial lung diseases (such as idiopathic pulmonary fibrosis), COVID‐19–related respiratory insufficiency, and Alzheimer’s disease. Its mechanism also suggests potential anti-inflammatory effects by modulating immune cell activity[1][3][5].
