Sennosides are a class of anthraquinone stimulant laxatives derived from the Senna plant (*Senna alexandrina*). They act as prodrugs that are converted by colonic bacteria into active rheinanthrones. These metabolites stimulate the myenteric plexus, leading to increased colonic motility and peristalsis, while also inhibiting water and electrolyte absorption and promoting their secretion into the intestinal lumen. In the clinical trial context of the COLONPREP study (NCT02744456) sponsored by Assistance Publique - Hôpitaux de Paris, sennosides are utilized as the mechanical bowel preparation (MBP) agent. The study evaluates the efficacy of sennosides-based MBP, alone or in combination with oral antibiotics, for the prevention of surgical site infections (SSI) in patients undergoing elective laparoscopic surgery for colon cancer.
