SENTI-202 is a logic-gated, allogeneic chimeric antigen receptor natural killer (CAR-NK) cell therapy developed by Senti Biosciences for the treatment of hematologic malignancies, primarily acute myeloid leukemia (AML) and myelodysplastic syndrome (MDS). The therapy uses engineered gene circuits to enhance selectivity and safety. It features an "OR gate" activating CAR that targets CD33 and FLT3 antigens on malignant cells, enabling the elimination of both leukemic blasts and stem cells. A "NOT gate" inhibitory CAR recognizes endomucin (EMCN) on healthy cells to prevent off-tumor toxicity. Additionally, calibrated-release IL-15 is included to improve persistence and activity of the NK cells. The product is manufactured from healthy adult donor NK cells that are cryopreserved prior to use[1][5][6][8].
