Overview

SENTI-301 is a multi-armed, off-the-shelf, healthy donor-derived chimeric antigen receptor natural killer (CAR-NK) cell therapy designed for the treatment of glypican 3 (GPC3) expressing tumors, particularly hepatocellular carcinoma (HCC). The engineered NK cells are modified to target the GPC3 antigen, which is highly expressed in most HCCs but has low or no expression on normal adult tissues. In addition to the anti-GPC3 CAR, SENTI-301 incorporates a proprietary calibrated release interleukin-15 (crIL-15) Gene Circuit. This payload is intended to stimulate surrounding immune cells and promote CAR-NK cell expansion, persistence, and tumor killing. The product is being developed by Senti Biosciences with clinical development and manufacturing in China led by Celest Therapeutics under the name SN301A[1][2][4][5][6].
