Senza5 CART5 is an autologous chimeric antigen receptor (CAR) T-cell therapy engineered to target the CD5 protein, which is expressed in approximately 85% of T-cell malignancies and certain B-cell lymphomas. A major challenge in targeting CD5 with T-cell therapies is "fratricide," where the CAR-T cells (which naturally express CD5) attack and kill each other. Senza5 CART5 overcomes this by utilizing CRISPR-Cas9 gene editing technology to knock out the endogenous CD5 gene in the patient's T cells before they are transduced with an anti-CD5 CAR. This modification prevents self-antigen recognition, thereby enhancing the expansion, persistence, and therapeutic durability of the CAR-T cells. The manufacturing process produces a product containing both CD5-knockout CAR-T cells and CD5-knockout untransduced T cells, the latter of which may help maintain immune function against pathogens while the CAR-T cells eliminate CD5-positive tumor cells.
